Paint.NET is a really slick paint program, and the best thing is that it's free. Layer and history support are great for us old-hat Photoshop users. I don't see it as a replacement for Photoshop, but it's definitely a replacement for Paint Shop Pro, which I hopefully won't ever have to install again. I've been installing Paint.NET on every computer that I can get my hands on because I feel that every computer needs a good image manipulation program and Microsoft Paint is totally lacking. I'd love it if Microsoft would work something out with the creators and make Paint.NET the replacement for Paint in future Windows releases, starting with Vista. Microsoft owes a better built-in paint program to its customers, in my opinion.
Edit: After watching the video, it sounds like Microsoft wants to replace Paint and suggested a new version of Paint to the creators. Perhaps there's more chance of seeing Paint.Net in Vista than I thought. I sure hope we see it included.
